چکیده

Some recent work, no tab ly tha t of Dixon and Elliott , 1 has indicated tha t not all cells and tissues are complete ly cyanide-sensit ive in the sense tha t they reduce their ra te of respiration in the presence of cyanides. Alien ~ has pointed out tha t not all the resp i ra tory exchange in Planaria m a y be inhibited b y cyanides. This has indicated tha t there m a y be other oxidation sys tems in the cells besides those affected by cyanides. Lund ~ found some years ago tha t K C N had litt le or no effect on oxygen consumption b y Paramecium. We proposed to reinvest igate this question and to determine if iron is present in the p ro top lasm of Paramecium, acting as the respirat ion ca ta lys t according to the theory of Warburg , and to note if addit ion of iron would cause an increase in the ra te of respiration in the Paramecium.

The Insensitivity of Paramecium to Cyanide and Effects of Iron on Respiration

1. The effects of KCN and iron salts on oxygen consumption has been studied in the cell of Paramecium caudatum by manometric methods. 2. KCN solutions of strengths from M/200 to M/10,000 have been shown to produce no decrease in oxygen consumption, but have in most cases produced a very slight increase in the respiration rate. 3. The pH values were found to have little or no effect on these res...

AMP-dependence of the cyanide-insensitive pathway in the respiratory chain of Paramecium tetraurelia.

The AMP-dependent stimulation of the cyanide-insensitive respiration of Paramecium mitochondria was investigated. The nucleotides exhibiting a stimulatory effect on the cyanide-insensitive oxidation of pyruvate (+ malate) in a medium supplemented with EDTA or carboxyatractyloside were, in decreasing order of efficiency, AMP, GMP, IMP, UMP and TMP. On the other hand, ADP, ATP and cyclic AMP were...

Occurrence of a para-nitrophenyl phosphate-phosphatase with calcineurin-like characteristics in Paramecium tetraurelia.

Using para-nitrophenyl phosphate (pNPP) as a substrate for enzymatic activity, we sought to identify CaN in Paramecium. We isolated three different pNPP-phosphatases from the soluble fraction of Paramecium cells by anion-exchange and affinity column chromatographies. One, pNPP-phosphatase Peak I, is very similar to mammalian CaN. Divalent cation dependency, inhibition by calmodulin (CaM) antago...

The secretory vesicle in living Paramecium is acidic.

In Paramecium, secretory proteins are packaged within membrane-bounded vesicles in a condensed form. This form expands when the proteins are released. We have now determined that a proton gradient is present in the secretory vesicles of living Paramecium. Acridine Orange, used as an in vivo indicator of acidic compartments, stained the secretory vesicles in both wild-type and mutant cells. Addi...

Interactions of membrane potential and cations in regulation of ciliary activity in Paramecium.

Ciliary activity in Paramecium was investigated in different external solutions using techniques of voltage clamp and high frequency cinematography. An increase in the external concentration of K, Ca or Mg ions decreased the resting potential. It had no effect on ciliary activity. When the membrane potential was fixed, an increase in external Ca or Mg and, to a lesser extent, an increase in K c...
